The Tetherin-Enabled Designer Cell Line for Next-Generation Vaccine Development

Expression of certain transgenes can interfere with the infectious cycle so that the parental viruses have a replication advantage. This situation can lead to transgene instability, and can emerge as a challenge in poxvirus vector development, limiting titers, compromising batch reproducibility, and forcing repeated plaque purification cycles. The AGE1.CR.ReX® host cell line solves this directly. Built on the proven GMP lineage of AGE1.CR.pIX®, it is equipped to stably express the Tet repressor (tetR) that allows transgene expression to be placed under inducible control during production. This eliminates the selection pressure against the challenging transgene. The result: higher titers, increased genetic stability, and production-ready virus in less time with greater confidence.

The AGE1.CR.ReX® host cell line is available in both adherent and suspension formats.

THE TECHNOLOGY BEHIND IT

The stably integrated tetR gene in AGE1.CR.ReX® is the foundation of the TetherexX System, our proprietary approach for rapid generation and rescue of pure recombinant MVA. Tetherin is a cellular factor of the innate immune system and used here as a novel selection marker. By controlling tetherin expression via TetR, the system selectively eliminates parental virus, delivering pure recombinant MVA in just three passages without plaque picking or operator-assisted steps.
